Depuis l'Antiquité, les rumeurs abondent sur des démons mangeurs d'hommes qui se cachent dans les bois. C'est pourquoi les habitants de la ville ne s'aventurent jamais dehors la nuit. Depuis la mort de son père, Kamado Tanjirou a pris sur lui de subvenir aux besoins de sa mère et de ses cinq frères et soeurs. Bien que leur vie soit rendue plus dure par la tragédie, ils ont trouvé le bonheur. Mais cette chaleur éphémère est brisée un jour lorsque Tanjirou découvre que sa famille a été massacrée et que la seule survivante, sa soeur Nezuko, s'est transformée en démon. Pour ajouter à cette tristesse, un chasseur de démons nommé Tomioka Giyuu arrive et est sur le point d'achever Nezuko, mais à sa grande surprise, elle et Tanjiro commencent à se protéger l'un l'autre. Voyant cette bizarrerie et les compétences de combat prometteuses de Tanjiro, Giyuu décide de les envoyer à son ancien mentor pour être formé. Ainsi commence la vie de Tanjiro en tant que chasseur de démons, lié à une quête pour guérir sa sœur et trouver celui qui a assassiné toute sa famille.
